Anti-TNF-alpha(1F6)抗體,腫瘤壞死因子-α單克隆抗體產品介紹：Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide stimulated macrophages, and causes tumor necrosis when injected into tumour bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five noncovalently linked units, but shows a single 17 kDa band following SDS PAGE under non reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumour Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis or cytostasis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

十二烷基硫酸鈉-聚丙烯酰胺凝膠電泳（SDS-PAGE）可用于檢測已被SDS化學變性的蛋白。然而，某些特定的抗體不會在一個變性的蛋白質分子上識別其抗原表位，這時需要進行不含SDS的聚丙烯酰胺凝膠電泳。

轉膜后，需對膜進行封閉，以減少非特異性蛋白結合，然后進行用一抗孵育膜，以研究感興趣的目標蛋白。單克隆抗體通常具有更高的特異性；然而許多單克隆抗體產生于某個特定肽段而不是一個天然蛋白質，因此可能無法檢測到PAGE所分離的天然蛋白。也可使用多克隆抗體，但易形成較高的背景值。因一抗通常無標記，所以需要一個可以結合在一抗Fc段的標記的二抗，以用于zui后的檢測。通常會用酶來標記二抗。酶標記后的印跡可通過化學發光酶底物及放射自顯影技術成像。被抗體檢測和標記到的蛋白質會出現在圖像的暗區。
