細胞周期檢測點激酶2抗體【背景介紹】：In response to DNA damage and replication blocks, cell cycle progression is halted through the control of critical cell cycle regulators. The protein encoded by this gene is a cell cycle checkpoint regulator and putative tumor suppressor. It contains a forkhead-associated protein interaction domain essential for activation in response to DNA damage and is rapidly phosphorylated in response to replication blocks and DNA damage. When activated, the encoded protein is known to inhibit CDC25C phosphatase, preventing entry into mitosis, and has been shown to stabilize the tumor suppressor protein p53, leading to cell cycle arrest in G1. In addition, this protein interacts with and phosphorylates BRCA1, allowing BRCA1 to restore survival after DNA damage. Mutations in this gene have been linked with Li-Fraumeni syndrome, a highly penetrant familial cancer phenotype usually associated with inherited mutations in TP53. Also, mutations in this gene are thought to confer a predisposition to sarcomas, breast cancer, and brain tumors. This nuclear protein is a member of the CDS1 subfamily of serine/threonine protein kinases. Several transcript variants encoding different isoforms have been found for this gene.

（一） 儀器設備
1. 18cm不銹鋼高壓鍋或電爐或用微波爐.
2. 水浴鍋
（二） 試 劑
1. PBS緩沖液（ph7.2―7.4）：NaC137mmol/L，KCl 2.7mmol/L ,Na2HPO4 4.3mmol/L,
KH2PO4 1.4mmol/L.
2．0.01mol/L檸檬酸鈉緩沖液（CB,ph6.0,1000ml）：檸檬酸三鈉3g,檸檬酸0.4g。
3．0.5mol/L EDTA緩沖液（ph8.0）：700ml水中溶解186.1g EDTA&
8226;2H2O,用10mmol/L
NaOH調至ph8.0,加水至1000ml.
4. 1mol/L的TBS緩沖液（ph8.0）：在800ml水中溶解121gTris堿，用1N的HCl調至ph8.0,加水1000ml。
5. 酶消化液：a. 0.1%*：用0.1%CaCl 12（ph7.8）配制。b.0.4%*液：用0.1N的HCl配制。
6. 3%甲醇―H2O2溶液：用30%H2O2和80%甲醇溶液配制
7. 風裱劑：a. 甘油和0.5mmol/L碳酸鹽緩沖液（ph9.0–9.5）等量混合
b 油和TBS（PBS）配制
8．TBS/PBS PH9.0–9.5,適用于熒光纖維鏡標本;ph7.0-7.4適合光學纖維標本。
（三） 操作流程
1 脫蠟和水化：脫蠟前應將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘。 a 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘
b 無水乙醇中浸泡五分鐘
c 95%乙醇中浸泡五分鐘
d 75%乙醇中浸泡五分鐘
2 抗原修復：用于福爾馬林固定的石蠟包埋組織芯片
